Burn Both Bootloader and Firmware!

You will load the bootloader first. The bootloader is so that you can load the FW using USB. Without the bootloader your computer USB doesn't recognize that there is a chip attached. You can also load the FW using the ISP so it's really your choice.

I have programmed the PIC successfully and my computer detects the Megadrum. Now I want to know If I can directly burn the firmware to the ATMEGA using the USBASP programmer without the bootloader?
